Course Curriculum
Note: This course focuses strictly on chromatographic separation, from manual basics to automated HPLC execution.
Module 1: Foundations, Safety & Manual Separation
- Introduction to Good Laboratory Practices (GLP) and essential lab safety.
- Principles of chromatography: stationary vs. mobile phases, partition coefficients, and polarity.
- Thin Layer Chromatography (TLC): solvent system selection, spotting, and calculating Rf values.
- Manual Column Chromatography: packing a silica gel column, loading samples, and fraction collection.
Module 2: HPLC Fundamentals & Crucial Sample Prep
- Transitioning to automated systems: understanding HPLC components (pumps, injectors, columns, detectors).
- Normal phase v/s reverse phase chromatography.
- Best practices for sample extraction, filtration, and dilution to prevent column degradation.
- Preparing and degassing HPLC-grade solvents and mobile phases.
Module 3: Hands-On HPLC Operation
- System startup, purging, and equilibration.
- Proper column installation and care.
- Setting up run parameters, flow rates, and injection volumes.
- Executing the run and monitoring baseline stability in real-time.
- Safe shutdown and column washing procedures.
Module 4: Data & Troubleshooting
- Reading and interpreting chromatograms (retention times, peak area, resolution).
- Identifying common issues: leaks, pressure spikes, and ghost peaks.
- Routine maintenance for prolonged instrument health.
