Course Curriculum
Note: This course focuses strictly on preparative separation and the operation of automated flash systems.
Module 1: Foundations, Safety & Method Development
- Introduction to Good Laboratory Practices (GLP) and essential lab safety.
- Principles of preparative chromatography and separation science.
- Thin Layer Chromatography (TLC): The foundation of flash method development.
- Calculating Rf values (ΔRf) and translating TLC solvent systems to Flash gradients.
Module 2: Flash System Fundamentals
- Anatomy of a Flash Chromatography system: pumps, UV detectors, and automated fraction collectors.
- Understanding stationary phases: normal phase (silica) vs. reverse phase (C18) cartridges.
- Selecting the right cartridge size based on sample mass and resolution requirements.
- Solvent selection, preparation, and managing mobile phase gradients.
Module 3: Hands-On Operation & Loading
- System startup, solvent priming, and cartridge equilibration.
- Sample introduction techniques: liquid loading vs. solid loading (dry loading).
- Programming the separation method, flow rates, and detector wavelengths.
- Executing the run and monitoring the real-time UV chromatogram.
Module 4: Post-Run Analysis & Troubleshooting
- Locating target compounds: mapping the chromatogram to collected fractions.
- TLC analysis of fractions to confirm purity before pooling.
- Troubleshooting common issues: over-pressure, poor peak resolution, and baseline drift.
- Proper system shutdown, column purging, and waste management.
